摘要
目的:观察前列腺素 E_1(PGE_1)对大鼠心肌缺血再灌注损伤的保护作用。方法:采用在体大鼠心肌缺血再灌注模型并以生理盐水为模型对照,用超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、丙二醛(MDA)试剂盒,以分光光度法测定再灌注心肌组织中 PGE_1组与模型对照组上述指标及 Ca^(2+)含量。结果:与模型对照组比较,PGE_1可显著提高再灌注心肌组织中 SOD、GSH-Px 活力(P<0.05~0.01);并能显著降低再灌注心肌组织中 MDA 及 Ca^(2+)含量(P<0.05~0.01)。结论:PGE_1保护心肌,减轻再灌注损伤的作用主要与其清除再灌注过程中自由基的功能有关。
Objective:To study the preventive effect of prostaglandin E_1 on myocardial ischemia reperfusion injury.Methods:In a myocardial ischemia reperfusion model induced by 30 min coronary oc- clusion and 60 min reperfusion in openchest anesthetized rats,malondialdehyde(MDA)and Ca^(2+)con- tents,superoxide dismutase(SOD)and glutathione peroxidase(GSH-Px)activities in myocardium were determined with spectrophotometric methods.Results:After myocardial ischemia reperfusion,an increase of MDA and Ca^(2+)contents,a decrease in SOD and GSH-Px activities were noted in reperfused myocardium. Prostaglandin E_1 effectively ameliorated the injury as manifested by decrease in Ca^(2+)and MDA contents( P<0.01)as well as increase in the SOD and GSH-Px activities(P<0.01).Conclusion:The preven- tive effect of prostaglandin E_1 against ischemia reperfusion injury may be due to scavenging the free radi- cals formed during reperfusion.
