摘要
以草菇Vol.0122为出发菌株,经过紫外线、紫外线和离子束复合诱变,结合双层平板分离技术选育出1株纤维素酶高产突变菌株TR12。确定培养基的最佳组成成分为(g/L):甜高粱秸秆粉15,麸皮粉15,(NH4)2SO43;最适产酶条件为:接种量2%(孢子悬液浓度3×107cfu/mL),pH值6.0,培养温度(32±1)℃,发酵时间120h,发酵罐转速120 ̄140r/min;所得粗酶液的CMC、FPA酶活分别为73.9U/mL和72.1U/mL。
Volvariella volvacea Vol.0122, a mutant stain with a high yield of cellulase is screened from an original stain Volvariella volvacea Vol12 by means of UV and ion beam mutagenisis together. The optimal components of the growth medium are (concentrations in g/L) wheat bran (15), corncob(15), (NH4)2SO4(3) and the optimal conditions yielding cellulase are 2% pitching rate(seed volume)(ie, 3×107 cfu/mL spore suspension), pH 6.0, temperatures (32±1)℃, fermentation of 120h, and 120~140r/min rotation speed. The cellulase activity of CMC and FPA are 73.9 U/mL and 72.1 U/mL respectively.
