摘要
目的建立一种实用的肺动脉高压动物模型。方法采用左肺叶切除复合皮下注射野百合碱(MCT)的方法。动物切除左肺后1周皮下注射MCT,注射后第1,2,3,4周分别测量平均肺动脉压(mPAP)和平均右室压(mPRV)后取其右肺、心脏,计算:(1)肺中型(外径50~150μm)和小型(15~50μm)肌型动脉的相对中膜厚度(RMT);(2)有完整内外两层弹力层的肌型动脉占肺小血管总数的百分比;(3)心脏称右室和左室加室间隔的质量,计算比值RV/(LV+S)。对照组仅行假手术,1周后皮下注射生理盐水。结果与对照组比较,左肺切除复合MCT注射后4周mPAP明显增高[(48.19±3.33)vs(17.21±1.26)mmHg,P<0.001],右心室肥厚明显[(0.93±0.02)vs(0.27±0.01),P<0.001];光镜下,肺小血管肌化程度增强,RMT明显增加[中型(30.38±1.29)%vs(7.98±0.48)%;小型(25.06±0.99)%vs(6.18±0.42)%,P<0.01],肺血管密度明显减少[(3.68±0.13)vs(7.65±0.25)个/mm2,P<0.01]。结论本方法建立的大鼠肺动脉高压模型是可行且实用的。
Objective To establish a practical animal model with pulmonary arterial hypertension (PAH). Methods Male SD rats underwent left pneumonectomy and then a hypodermal injection of monocrotaline(MCT) at a dose of 60 mg/kg. At different intervals of 1, 2, 3 and 4 weeks after MCT injection, the rats were measured of homodynamic and harvested of their right lungs and hearts. Then (1) relative thickness of tunica media of pulmonary muscularized arterioles, (2)percentage of muscular arterioles with intact elastic layers amongst the pulmonary vessels, and, (3)ratio of right ventricle(RV) to left ventricle(LV) +ventricle septum(S) were calculated respectively for evaluation of pulmonary vascular remodeling. The controls just underwent a sham operation and then an injection of normal saline. Results Cardiac hypertrophy and symptom of right heart failure such as hydrothorax, ascites and congestive liver cirrhosis were observed in the end-staged PAH rats. In comparison to the controls, mPAP and RV/(LV +S) were markly elevated [(48.19±3.33) vs (17.21±1. 26)mmHg]; (0.93±0.02) vs (0.27±0.01), P 〈0.001, the density of pulmonary vessels was greatly reduced[(3.68±0.13) vs (7.65±0.25)mm^-2, P〈 0. 01], that indicating a well formation of PAH. In addition, muscularization of pulmonary arterioles as well as hypertrophy of tunica media was observed in the right lung specimens. Conclusion Rat model with PAH established by left pneumonectomy and MCT injection is a useful and practical one for further study of PAH.
