摘要
目的:观察归知糖疽颗粒对实验性糖尿病大鼠皮肤溃疡模型创面愈合率、病理组织学、血糖及小鼠微循环障碍的影响。方法:1.链脲佐菌素(STZ)腹腔注射造成大鼠糖尿病模型再以外科手术方法制作皮肤损伤;用医学病理图象分析仪计算创面愈合率,用血糖检测仪检测血糖值,创面皮肤常规HE染色光镜观察病理变化。2.末次给药后30 min经尾静脉注射高分子右旋糖酐诱发小鼠耳廓微循环障碍模型,在显微镜下观察不同时段血液流态的改变。结果:归知糖疽颗粒0.81,1.62,3.24 g·kg^-1组治疗性灌胃给药4周糖尿病大鼠皮肤溃疡模型创面愈合率明显升高,分别为88.55%、89.49%和89.69%(均P〈0.001);血糖值明显降低(均P〈0.05);改善溃疡创面病理改变,胶原纤维细胞增多,炎性细胞减少。归知糖疽颗粒灌胃给药3d,1.17 g.kg,4.68 g·kg^-1组在20 min,30 min时段明显改善小鼠耳廓微循环障碍的血液流态(P〈0.05~0.01)。结论:归知糖疽颗粒有促进糖尿病大鼠皮肤溃疡模型皮肤溃疡创面愈合、降低血糖、改善微循环障碍作用。
Objective: To study the effect of Guizhi Tangju Granules on blood sugar, dermal ulcer in streptozotocin-induced diabetic rats and effects on microcirculation in mouse. Methods: 1. Wistar rats injected with streptozotocin(STZ) ip, at the same time, skin destruction made by surgical intervention. Healing ration and pathological change of dermal ulcer were observed. 2. Microcirculation disturbance induced by injection of dextran intravenously was observed after mouse were fed with Guizhi Tangju Granules for 4 weeks. Results: Guizhi Tangju Granules could significently improve microcirculation disturbance in mouse and reduce blood sugar, increase healing ration of damaged skin in streptozotocin-induced diabetic rats, as well as raise collagenous fibers cells and decrease inflammatory cells in pathology. Conclusion: Guizhi Tangju Granules could promote healing of damaged skin, decrease serum glucose and improve microcirculation disturbance.
