摘要
介绍了不同孔径(10 nm、100 nm、300nm)的硅胶聚合物键合相的合成,比较了其对蛋白质的分离特性。结果表明,大孔径的硅胶聚合物键合相比表面积小,易于获得生物大分子的快速分离。探讨了在孔径为 300 nm的硅胶聚合物键合相上,流动相的pH值及盐浓度对蛋白质保留时间的影响。
The preparation of macroporous silica gel matrices of different pore diameter, the synthesis of different pore diameter silica-based ploymer-bonded phaese and their chro- matographic characteristics for the separation of proteins are introduced. The results ob- trained show that macroporous packings have relatively small surface area and low capacity, and therefore benefit for the quick separation and analysis of biopolymers. The effect of salt concentration and pH value of the eluent on the retention of proteins on 300 nm silica-based polymer-bonded phase is also discussed.
