摘要
选择PEG-柠檬酸钠双水相体系对AS1.398中性蛋白酶粗酶液进行萃取纯化研究,考察粗酶质量浓度、pH值、相比、离子强度等因素对酶分配的影响,并进一步采用响应面分析优化实验条件.结果表明:采用响应面方法可以有效地对系统进行选择和设计,大大方便了研究工作.选择PEG1000-柠檬酸钠体系,在粗酶质量浓度5.2 g/L、相比2、pH=7.1条件下,AS1.398中性蛋白酶的纯化倍数达3.6以上.
In this paper, PEG-citrate aqueous two-phase systems were chosen to purify AS1. 398 neutral protease. The effects of PEG molecular weight, mass concentration of raw enzyme, pH value, phase volume ratio and NaCl addition on the partition of AS1. 398 neutral protease were investigated. Response surface methodology was employed to further analyze and optimize the purification of AS1. 398 neutral protease. Research indicates that for the PEG1000/citric acid sodium system,when phase volume ratio = 2, pH value = 7.1, p(raw enzyme) = 5.2 g/L, the purification coefficient is more than 3.6 folds.
