摘要
目的:探讨保肝宁抗肝纤维化的作用机制。方法:采用复合因素建立肝纤维化大鼠模型,采用全自动生化分析仪检测谷丙转氨酶(ALT)、谷草转氨酶(AST),总胆固醇(TC)、甘油三脂(TG),用免疫组化方法观察肝纤维化大鼠肝组织瘦素(Leptin)表达水平。观察保肝宁对肝纤维化模型大鼠ALT、AST、AST/ALT,TC、TG及大鼠肝组织Leptin表达水平等各项指标的影响。结果:与正常组比较,模型组及各药物组血清ALT、AST、AST/ALT比值均明显升高(P均<0.01)。与模型组比较,保肝宁药物组血清ALT、AST明显下降,差异有显著性意义(P均<0.01),保肝宁大剂量降低AST作用明显优于秋水仙碱(P<0.05)。与模型组比较,保肝宁大、中剂量药物组血清TC、TG水平明显升高,差异有非常显著性意义(P均<0.01)。保肝宁明显抑制模型大鼠肝组织Leptin的表达。与模型组比较,差异有显著性意义(P均<0.01)。结论:保肝宁可有效防治大鼠肝纤维化,其作用机制可能与通过改善肝功能,调节血脂及抑制模型大鼠肝组织Leptin的表达密切相关。
Objective:Discuss the mechanism of Baoganning against liver fibrosis.Methods:Adopt complex factors to set up fibrosis rat model,then employ automatic biochemical analyzer to detect ALT、AST、TC、TG,and immunohistochemical methods to detect the expression level of leptin in the liver tissue of model rats.Observe the influence of Baoganning on ALT、AST、AST/ALT 、TC、TG,and leptin level of liver fibrosis rat model.Results:Compared with normal group,the indexes as serum ALT、AST、AST/ALT were apparently increased in model group and every drug group(P<0.01 in all).According to model group,the indexes as ALT、AST and leptin level were obviously dropped in drug group using Baoganning,with notable significance in their differences(P<0.01 in all).The effect of lowering AST with large dose of Baoganning was evidently better than that with colchicines(P<0.05).Compared with model group,serum TC、TG level increased apparently with large and medium dose of Baoganning in drug group,with very notable significance(P<0.01 in all).Baoganning can obviously restrain the expression of leptin in the liver tissue of model rats,with notable significance in their differences(P<0.01 in all).Conclusion:Baoganning can effectively prevent and treat liver fibrosis in rats,whose mechanism may have a close connection with the improvement of liver function,the adjustment of blood lipid and the restraint of the expression of leptin in liver tissue of model rat.
