摘要 采用重氮化法,将磺胺间甲嘧啶(SM1)与牛血清蛋白(BSA)偶联制备合成抗原BSA-SM1,并用作免疫抗原,用紫外扫描法和聚丙烯酰胺电泳法对其进行鉴定,并测得偶联的结合比。结果表明,通过紫外扫描和聚丙烯酰胺电泳法的测定证明偶联成功,偶联的结合比为9∶1。用BSA-SDM免疫BALB/C小鼠,间接ELISA测定多抗上清(pAb)效价,获得了高效价的pAb,为SM1残留免疫学检测方法的建立奠定了基础。 In this paper, Bovine serum albumin (BSA) was used as a protein carrier to couple with semiantigen Sulfamerazine (SM1) by diazotization method. The complete antigen BSA-SM was thus prepared as immunoantigen in enzyme-linked immunosorbent assay (ELISA) protocol. UV spectrophoto metry and Sodium dodecyl-sulfatepolycrylamide gel electrophonesis (SDS-PAGE) were applied to determine it. And the conjugated ratio was estimated. The results indicated that the conjugation was succeed and the conjugated ratio of SM1 and BSA was 9:1. The high-titer anti-SM1 pAb was been produced in serum of immunized BALB/C mice, which made it possible to establish method of SM1 residues in feed and animal food.
