摘要
利用本实验室cDNA文库测序所获得的茶树EST序列为主自行开发的24对和他人7对共31对EST-SSR引物,对60份西南茶区茶树资源进行了遗传多样性和亲缘关系分析。结果表明,西南茶区茶树资源的遗传多样性非常丰富。31对引物共检测到等位位点137个,每个引物为2 ̄8个之间,平均4.42个。遗传多态性信息含量(PIC)在0.09 ̄0.85之间,平均达0.63。观测杂合度(Ho)在0.07 ̄0.94之间,平均值为0.53。UPGMA聚类表明,按相似系数为0.39可将参试的60份种质资源分为五大类。聚类结果在分子水平上显示了我国西南茶区种质资源间的亲缘关系,为以后种质资源保存和新品种选育提供一定的启示。
In total, 31 pairs of EST-SSR primers, among them 24 were newly developed from the ESTs of tea plant and 7 were designed from references, were used to analyze the genetic polymorphism and genetic relationship of 60 tea germplasms from Tea Area in Southwest China. The results showed that the genetic polymorphism of tea germplasms in the Tea Area in Southwest China were very abundant. Totally 137 alleles were amplified using 31 EST-SSR primers, the number of alleles per primer ranged from 2 to 8, on average of 4.42. The polymorphism informarion content (PIC) varied from 0.09 to 0.85 and the observed heterozygosity (Ho) ranged from 0.07 to 0.94, on average of 0.63 and 0.53, respectively. The 60 accessions were classified into 5 groups based on the UPGMA method with the coefficient at 0.39. The genetic relationship of all 60 accessions revealed by the EST-SSR analysis would not only be of great help to conserve the tea germplasms but also give some useful implications for tea breeding,
