摘要
目的通过诱导檀香体细胞胚胎(体胚)发生获得再生植株,为檀香的基因工程提供受体系统。方法分别以檀香幼嫩叶片、茎尖和近成熟胚为外植体,在添加TDZ和其它植物生长调节剂的MS培养基上诱导体胚的发生,再将发育成子叶型的体胚转移到含赤霉素(GA3)的培养基上萌发成完整植株。结果叶片外植体在附加1.1μmol/LTDZ的MS培养基最适于体胚的发生,体胚发生率为91.2%;体胚在附加1.4μmol/LGA3,蔗糖含量为4%的MS培养基上的萌发率达100%,但形成的檀香幼苗未见根生长。结论1.1μmmol/L的TDZ能诱导檀香叶片和近成熟的种子胚外植体形成体胚,体胚在附加1.4μmol/LGA3的MS培养基上能萌发,但萌发的幼苗没有生长出正常的根。
Objective To obtain the regenerated plantlet of Sandalwood (Santalum album L.) through somatic embryogenesis and establish a genetic transformation system for future experiments. Methods Using the shoot tips, leaves of top 3 nodes and the nearly mature embryos as explants, somatic embryogenesis was induced on Murashige and Skoog (MS) medium containing thidiazuron(TDZ) and other exogenous growth regulators, and somatic embryos were isolated and transferred to MS medium with GA3 to develope into whole plants. Results The explants from leaves was most suitable to form somatic embryos on MS medium containing 1.1 μmmol / L TDZ and the formation rate of somatic embryos was 91.2%. The somatic embryos were developed into rootless plantlets by reculturing onto MS medium with 1.4 μmol / L GA3 and 4% sucrose, and the germination rate was 100 %. Conclusion 1.1 μmol / L TDZ can induce the somatic embryogenesis from leaves and nearly mature embryos of sandalwood, and the somatic embryos can be develope into rootless plantlets on MS medium with 1.4 μmol / L GA3.
