电针对慢性应激抑郁模型大鼠海马糖皮质激素受体和N-甲基-D-天冬氨酸受体2B表达的影响 被引量:6

Influence of Electroacupuncture on the Expression of GR and NR2B in Hippocampal of Chronic Stress Depression Model Rats

摘要 目的探讨电针对慢性应激模型大鼠抑郁状态相关行为学的影响及其作用机制。方法40只SD大鼠分为空白组、模型组、电针组和氟西汀组,应用慢性复合应激方法造模,进行抑郁状态相关行为学检测,包括Open-field实验、强迫游泳实验和"Y"迷宫实验。免疫组化方法检测海马内糖皮质激素受体(GR)、N-甲基-D-天冬氨酸受体(NR)2B的表达情况。结果与空白组比较,模型组大鼠的探究活动减少(P<0.01),学习记忆能力降低(P<0.01),电针组大鼠相应行为学实验比模型组有显著提高(P<0.01);模型大鼠海马内GR、NR2B阳性表达强度显著降低(P<0.01),电针组海马内NR2B的阳性表达强度有所提高(P<0.01),而GR也表现出升高的趋势。结论电针可以减轻慢性应激引起大鼠抑郁状态的程度,其机制可能与脑内GR、NR2B表达变化有关。 Objective To investigate the influence of electroacupuncture (EA) on behaviors and GR, NR2B in hippocampal of the chronic stress depression rats. Methods Forty SD rats were divided into control, model, EA and Prozac groups. Chronic stress model was established by using chronic mixed stimulation. Observe the behaviors caused by chronic stress, including changes of spontaneous movements in Open-field test, learning-memory behavior in“Y”-maze test and stillness time in forced-swimming test. The expression of GR and NR2B in hippocampal tissue was detected by immunohistochemical technology. Results Compared with control group, the results of spontaneous movements, stillness swimming time and learning-memory behaviors of the chronic stress model rats were declined significantly (P 〈0.01), which of EA group were improved compared with model group (P〈0.01). The expression of GR and NR2B in hippocampal of model rats was declined significantly compared with control group (P〈0.01), and NR2B was increased in EA group (P〈0.05, P〈0.01), GR also showed the intend to be increased. Conclusion EA could reduce the changes of behaviors which were caused by chronic stress, the possible mechanism of it might related to the changes of expression of GR and NR2B in the hippocampl.
