摘要
[目的]为商城肥鲵的生物学研究和两栖类血液生理比较细胞学研究提供基础资料。[方法]采集商城肥鲵的血液,用常规法制成血涂片。用姬姆萨染液染色后,对商城肥鲵血细胞形态进行观察,并对其血液进行检验分析。[结果]在商城肥鲵血涂片中可鉴定出红细胞、单核细胞、淋巴细胞、血栓细胞、嗜酸性粒细胞、嗜碱性粒细胞和嗜中性粒细胞。红细胞数量较多,长椭圆形,具圆核。白细胞数量少,圆形,其中淋巴细胞和单核细胞含量最高。商城肥鲵的红细胞平均数为3.2×105/mm3,白细胞平均数为9.1×104/mm3,血红蛋白含量为48 g/L,凝血时间为6.2 min。[结论]商城肥鲵红细胞大小平均为15.5μm×9.6μm,比东北小鲵小,而红细胞数量却比东北小鲵多,这可能与其在两栖类中所处位置有关,并与其生态习性相适应。
[ Objective] The research aimed to provide basic data for biology research on Pachyhynobius shangchengensis and comparative cytology research of blood physiology in amphibian. [Method] The blood of P. shangchengensis was collected to make into blood smear by using conventional method. After being dyed with Giemsa＇ s dye, the morphology of blood ceils in P. shangchengensis was observed and its blood was tested. [ Result ] From the blood smear of P. shangchengensis, erythrocytes, monocytes, lymphocytes, thrombocytes, eosinophilic granulocytes, basophilic granulocytes and neutrophillc granulocytes were identified. The amount of erythrocytes were more and they were lang elliptical with round nucleus. While the amount of leucocytes was small and they were rounded, in which the content of lymphocytes and monocytes was highest. The average erythrocytes in P. shangchengensis was 3.2 × 10^5/mm^3,the average leucocytes was 9.11 × 10^4/mm^3,the content of haemoglobin was 48 g/L and the blood coagulation time was 6.2 rain. [Conclusion]The average size of erythrocyte in P. shangchengensis was 15.5 μm × 9.6 μm, smaller than that of Hynobius leechii, while its amount of erythrocytes was more than that of H. leechii, which was possibly related with its position in amphibian and it was be suitable for its ecological habits.
