摘要
采用正交试验设计方法研究了MS微量元素,2,4-D、萘乙酸、脯氨酸和腺嘌呤的浓度对5种基因型大葱未成熟子叶愈伤形成和体细胞胚胎发生的影响。结果表明,MS微量元素的浓度对出愈率和胚胎发生率均有显著的作用;在MS培养基中附加2,4-D、萘乙酸、脯氨酸和腺嘌呤对愈伤组织的形成有明显的促进作用。微量元素浓度对大葱体细胞发生有明显的影响,各基因型间在胚胎发生时所需激素种类和浓度存在明显差异。
The effects of the concentration of MS micro salts,2,4-dichlorophenoxyacetic acd(2,4-D),naphthalene acetic acid(NAA),prolin and adenine on callus formation and embryogenesis were investigated by using orthogonal design with immature cotyledon of Chinese onion.The results showed that the role of concentration of NAA,proline or adenine could promote callus formation.The concentration of MS mecro salts possessed obvious effects on somatic enbryogenesis of Chiese onion.The category and concentration of the hormone needed were different among genotypes when embryogenesis was induced.
