摘要
用MT+BA(10mg/L)+5%蔗糖诱导锦橙幼龄受精与未受精胚珠发生珠心愈伤组织,转入MT+5%半乳糖后促进了珠心愈伤组织的高频体胚发生。用聚丙烯酰胺凝胶等电聚焦研究珠心愈伤组织胚发生早期过氧化物酶(POD)同工酶的表达,发现在固体培养和悬浮培养方式中不同的分化培养基上,球形胚及球形胚形成前10天的培养物都存在一个分化培养前的胚性愈伤组织所没有的酸性POD同工酶。
Nucellar callus was induced from young fertilized and unfertilized ovules of “Jincheng” orange (Citrus sinensis Osb) on the solid MT medium supplemented with 10mg/L BA Somatic embryogenesis at a high frequency in nucellar callus was induced on the solid MT medium containing 5% galactose Developmental expression of peroxidase (POD) isozymes during early stages of embryogenesis in nucellar callus was studied by polyacrylamide gel isoelectric focusing Comparison of POD isozyme patterns showed there apparently existed a specific acidic POD isozyme pattern before the formation of globular stage embryos Moreover, heavy expression of the POD isozymes couldn′t be effected by different culture methods —— solid culture and liquid suspension culture, with different embryo-inducing media The preliminary result showed the stage —— specific acidic isoperoxidase as a biochemical marker of early embryogenesis in citrus nucellar callus
