摘要
目的观察氯沙坦对颈总动脉损伤后再狭窄大鼠血浆内皮素(ET)和颈总动脉平滑肌细胞(SMC)中血小板衍生生长因子(PDGF)表达的影响。方法采用大鼠颈总动脉挤压术制备再狭窄模型,设假手术组、手术组和氯沙坦组,每组10只。观察颈总动脉形态学变化,放射免疫法测定血浆ET含量,免疫组织化学法观察颈总动脉PDGF-B的表达。结果假手术组颈总动脉各层结构正常、清晰,无明显血管平滑肌细胞增殖;手术组大鼠颈总动脉SMC大量增殖,内膜显著增厚;氯沙坦组SMC增殖程度降低。与假手术组比较,手术组大鼠血浆ET含量显著升高(P<0.01),颈总动脉SMC中PDGF-B表达显著增强(P<0.01);与手术组比较,氯沙坦组大鼠血浆ET含量与PDGF-B表达显著下降(P<0.05)。结论ET、PDGF参与血管再狭窄的形成,氯沙坦可通过降低ET含量与PDGF-B表达实现抗再狭窄的作用。
Objective To observe the effects of losartan on endothlin(ET) level and expression of platelet-derived growth factor in rats with carotid artery restenosis. Methods Restenosis model was established. Treatment with losartan as losartan group. The morphologie change and level of ET were measured. The expression of PDGF-B was detected by immunohistoehemieal technique. Results Compared with sham group,the sections of injured carotid artery showed intimal thickening with large numbers of VSMCs proliferating,the level of ET was obviously increased( P 〈 0.01 ) ,the expression of PDGF-B was significantly increased( P 〈0.01 )at 14th day after operation. Compared with operating group, VSMCs proliferating was inhibited and also the ET level, expression of PDGF-B were decreased obviously in losartan group (P 〈 0.05, respectively). Conclusion ET, PDGF-B take part in the vascular restenosis, Losartan is to be the therapy for vascular restenosis through inhibiting ET level and PDGF-B expression.
