摘要
目的探讨NF-κB在胆管癌中的表达及与胆管癌发生发展的关系。方法采用免疫组织化学方法对56例胆管癌和37例良性病变肝外胆管组织中NF-κBp65、cyclinD1、VEGF的表达进行检测,并对其与临床病理因素之间的关系进行分析。结果NF-κBp65、cyclinD1、VEGF在胆管癌和良性病变肝外胆管组之间差异有统计学意义(P<0.01)。NF-κBp65的表达与性别、年龄、肿块部位无关(P>0.05),而与组织分化、有无淋巴转移和神经浸润有关(P<0.01)。NF-κBp65与cyclinD1、VEGF的相关系数分别为0.590、0.683,有明显的相关性。结论NF-κBp65在胆管癌中高表达,并通过上调cyclinD1及VEGF的表达,促进胆管癌的发生、发展、浸润、转移。
Objective To investigate NF-κB expression and its possible mechanisms in the development of bile duct carcinoma.Methods The expression of NF-κBp65,cyclinD1 and VEGF was determined by Power Vision immunohistochemical analysis in 56 cases of bile duct carcinoma and 37 cases of normal bile duct.Results The positive rates of NF-κBp65,cyclinD1 and VEGF were significantly higher in bile duct carcinoma than in normal bile duct tissue(P〈0.01).The expression of NF-κBp65 was not correlated with age,sex and mass location(P〉0.05),but correlated with histological type(P〈0.01).The expression of NF-κB had significant difference between patients with positive and negative lymph node metastasis,also between patients with positive and negative nerve invasion(P〈0.01).NF-κB was correlated with cyclinD1 and VEGF(r=0.590,r=0.683,P〈0.01).Conclusion The results suggest that NF-κB could play an important role in the occurrence,development,invasion and metastasis of bile duct carcinoma by up-regulating cyclinD1 and VEGF expression.
