兔高眼压模型的眼球超微结构改变 被引量:3

Ultrastructure of rabbit model of high intraocular pressure

摘要 目的观察复方卡波姆诱导的兔慢性高眼压模型参数的改变,研究慢性眼压升高对兔眼球结构的损害。方法将32只青紫兰兔随机分为8组,左眼前房内注射0.3%复方卡波姆0.3mL,术后1、2、3、4、6、8、10、12周分别处死1组兔,制作眼球标本、视网膜组织悬液,图像分析技术定量进行分析。结果随药物诱导时间的延长,眼压升高持续约3个月,平均眼压为27~38mmHg,模型成功率为95.7%。视网膜各层显著变薄,视神经节细胞(RGCs)密度和视神经纤维层(RNFL)厚度减少率分别为57.8%和66.7%。视网膜神经细胞死亡的主要形式是凋亡,坏死细胞的比例随高眼压的作用无明显变化。结论复方卡波姆诱导的兔青光眼模型具有引起眼压中度、稳定、缓慢升高且时间长和可控制等优点,引起的高眼压性眼底损害和人类青光眼的眼底损害类似。 Objective High intraocular pressure (lOP) directly leads to the optic nerve atrophy and therefore loss of visual function. This study was to observe the relavant parameter changes of chronic high lOP model induced by compound carbomer in rabbits and study the ultrastructure change of chronic high lOP eyes. Methods The chronic hypertension model was established in the left eyes of 32 healthy pigmented rabbits by the injection of O. 3 mL of O. 3% carbomer into anterior chamber,and the PBS was injected at the same way in the right eyes. The model eyes were randomly divided into 8 groups according to the different treating time. The experimental rabbits were sacrificed in 1 week,2,3,4,6,8,10 and 12 weeks for the histological examination under the electron microscope. Retinal single cell suspension was taken and flow cytometry assay was used for the quantitative analysis of apoptosis and necrosis. Results With drug-induced extension of time,the slow increase of lOP,retinal thickness,increased neuronal cell death were found. High lOP lasted about 3 months. The average lOP was 27 -38 mmHg with the top lOP 37 -45 mmHg. The successful rate of glaucoma model was 95.7%. The various layers of retina showed the different degree of significantly thinner, and the density of retinal ganglion ceils(RGCs) and the thickness of retinal nerve fiber layer(RNFL) reduced by 57.8% and 66.7% in the hypertention eyes. The main form of retinal nerve ceils death was apoptosis,and no significant change was seen in the proportion of ceils necrosis in high lOP eyes. Conclusion Compound carbomer induces moderate, stable, slow and lastlng-time hypertention in rabbits with a similar pathologic characteristics in optical nerve and retina of human glaucoma.
