摘要
目的:探讨重楼总皂苷对热灭活大肠杆菌诱导大鼠腹腔巨噬细胞(PMΦ)分泌TNF-α、IL-1β的影响。方法:分离并纯化大鼠腹腔巨噬细胞(PMΦ),与重楼总皂苷共同孵育,2h后予热灭活大肠杆菌(终浓度3.5×107CFU/ml)诱导PMΦ释放TNF-α、IL-1β,以ELISA酶联免疫吸附测定法检测上清液中TNF-α、IL-1β浓度。结果:重楼总皂苷对热灭活大肠杆菌诱导PMΦ释放TNF-α、IL-1β具有显著的抑制作用(P<0.01)。结论:重楼总皂苷可以抑制腹腔巨噬细胞分泌TNF-α、IL-1β的活性。
objective: The aim is to study the effect of Rhizoma Paridis total saponins on TNF-α and IL-1β secretion in rat peri- toneal macrophages induced by heat-inactivated eseherichia coli strain. Methods: The rat peritoneal macrophages was isolated and puri- fied, being cultivated with Rhizoma Paridis total saponins. 2 hours later, heat-inactivated escherichia coli strain (3.5 × 10^CFU/ml) was given to increase the releasing of TNF-α and IL-1β nduced by PMdp. The concentration of TNF-α and IL-1β in supernatant was determined by ELISA. Results: The heat-inactivated escherichia coli strain induced secretion of TNF-α and IL-1β in PMdp could be significantly inhibited by Rhizoma Paridis total saponins ( P 〈 0.01 ).
