摘要 C-反应蛋白(C-reactiveprotein,CRP)是一种肝脏合成的急性时相蛋白,由5个相似的大小约为23kDa的亚基以非共价键相连,均衡对称分布。具有这种蛋白结构的相关蛋白同属于五聚体血浆蛋白家族(penraxin)。CRP参与机体对炎症的免疫反应,包括在Ca^(2+)存在下和胆碱磷酸结合来识别病原体,通过和Clq直接结合而激活补体经典途径,还可以和免疫球蛋白的受体FcγRI和FcγRⅡ结合,诱导嗜菌作用的发生。在炎症期,CRP的血浆浓度迅速上升,这一特性在医学上被广泛应用,近几年来,水产养殖业也利用CRP的这一特性来监测鱼类的健康状况。目前,在无脊椎动物鲎和许多脊椎动物中都已经发现了CRP的存在。CRP在系统演化中高度保守,是pentraxin家族的典型代表,起源于古老的五聚体血浆蛋白基因的基因倍增。本文综述了CRP的结构特征,在系统进化中的意义及其在临床实践和水产养殖中的应用。 C-reactive protein (CRP) is a acute-phase protein synthesized in the liver, consisting of five identical, noncovalently associated -23kDa protomers arranged symmetrically. The term "pentraxin" has been used to describe the family of related proteins with this structure. CRP participates in the systemic response to inflammation, including calcium dependent phosphocholine-binding to recognize pathogens, activating the classical complement pathway through direct interaction with C 1 q, interacting with the immunoglobulin receptors Fc,y RI and Fc,y RII eliciting a response from phagocytic cells. Its rapid increase in synthesis during inflammatory states is largely used for medical purpose. Furthermore, the healthy status of fishes in the aquiculture can be supervised with the characteristic of CRP during these years. CRP is a phylogenetically highly conserved plasma protein found from invertebrate limulus to mammals. It is a classical member of pentraxin family proteins which arose from a gene duplication of an ancestral pentraxin gene. This paper gave a general and detail overview on the structural characters, the significances in evolution and the applications in the clinical and aquiculture fields of C-reactive protein.
