摘要
目的:观察不同穴位处方电针对失眠大鼠下丘脑白细胞介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)质量浓度的影响,探讨电针治疗失眠症的作用机理。方法:将SD大鼠36只随机分为6组,即对照组、模型组、安定组、电针1组(取百会、神道、心俞穴)、电针2组(取三阴交、神门穴)、电针3组(取申脉、照海穴),模型组以腹腔注射对氯苯丙氨酸(PCPA)建立大鼠失眠模型,安定组给予腹腔注射安定,电针各组分别按穴位给予电针治疗,每组连续治疗6 d后检测大鼠下丘脑细胞因子IL-1β、TNF-α及IL-6的变化。结果:电针各组IL-1β、TNF-α和IL-6明显升高,与模型组和安定组比较差异显著(P<0.05,P<0.01);电针各组之间比较,电针2组好于电针1组(P<0.05),电针3组与电针1组、电针2组比较均无显著性差异(P>0.05)。结论:电针可能是通过提高失眠大鼠下丘脑细胞因子IL-1β、TNF-α和IL-6的质量浓度而改善睡眠;电针2组作用好于电针1组,说明取三阴交、神门穴治疗失眠症疗效更好。
Aim:To observe the effects of Electroacupuncture at different acupoints group on the content of Interleukin-1 β( IL-1 β ), Tumor Necrosis Factor- α( TNF-α), Interleukin 6 ( IL-6 ) in the hypothalamus of insomnia rats to investigate the mechanism of Electroacupuncture treatment of insomnia. Methods: Thirty-six SD rats were randomly divided into six groups: control group, model group, diazepam group, Electroacupuncture group I ( Baihui ( DU20 ), Shendao ( DU11 ), Xinshu ( BL15 ) ), Electroacupuncture group II ( Sanyinjiao ( SP6 ), Shenmen ( HT7 ) ) , Electroacupuncture group III ( Shemnai (BL62), Zhaohai (KI6)). The model of insomnia rats were established by DL-P-Chlorophenylalanine (PCPA) peritoneal injection. The diazepam group rats were treated by diazepam peritoneal injection. Each Electroacupuncture group rats were treated by Electroacupuncture at different acupoints. Cytokinessuch as IL-1β ,TNF-α ,IL-6 in the hypothalamus of insomnia rats in each group were determined after 6 days continuously treatment. Results: After Electroacupuncture treatment the content of IL-1β, TNF-α and IL-6 mong the no signifi increased obviously in contrast with model group and diazepam group(P 〈0. 05, P 〈0. 01 ). Athree Electroacupuncture groups, groupII was better than the group I(P 〈0. 05) ,but there was cant difference between the group III and the other two group (P 〉 0. 05). Conclusion:The mechanism of Electroacupuncture treatment of insomnia may be due to the increase of cytokines such as IL-1β TNF-α, IL-6 in the hypothalamus of insomnia rats. Electroacupuncture group II is better than group I and it may be suggest that Sanyinjiao(SP6), Shenmen(HT7) is better than other acupoints in treating insomnia.
