摘要
目的:建立人工绝经期模型,在降低雌激素水平的状态下,观察卵巢切除对睑板腺腺泡上皮细胞脂质的影响。方法:实验于2006-08/2007-05在河北工程大学医学院人体解剖学与组织胚胎学教研室完成。①实验材料:56~60d SD雌性大鼠72只,体质量200~240g;苏丹Ⅲ为天津科密欧化学试剂厂产品。②实验分组及过程:随机对72只青春期SD雌性大鼠分为3组:手术组、假手术组和正常组,每组24只,分别饲养2,4,6,8周后,用2%荧光素钠滴入大鼠结膜囊内,瞬目后在裂隙灯显微镜下用钴蓝光扫描照射观测大鼠的泪膜破裂时间,然后麻醉下颈椎脱臼法处死大鼠。③实验评估:采用放射免疫分析方法检测血清雌二醇水平;采用组织化学苏丹Ⅲ染色方法对睑板腺腺泡上皮做脂质染色阳性和强度检测。实验过程中对动物处置符合动物伦理学标准。结果:①手术组随术后时间的延长血清雌二醇水平逐渐降低,术后第4,6,8周低于正常组和假手术组(分别P〈0.05,P〈0.01,P〈0.01)。②手术组泪膜破裂时间于术后第4,6,8周明显缩短,与正常组、假手术组之间比较差异有显著性(P值均〈0.05),③手术组苏丹Ⅲ染色强度随时间延长逐渐减弱,术后第6周、第8周与正常组和假手术组之间比较差异有统计学意义(分别为P〈0.05,P〈0.01)。④血清雌二醇水平与泪膜破裂时间、苏丹Ⅲ染色强度有显著性相关关系(相关系数分别为r=0.9957,P=0.0043;r=0.9723,P=0.0036)。结论:卵巢切除后血清雌二醇水平降低,可造成睑板腺腺泡上皮细胞脂质减少,是导致睑板腺机能障碍的原因之一。
AIM: To establish a model of artificial menopause in ovariectomized rats, and observe the effect of ovariectomy on the lipids of meibomian gland acinar cells under decreased serum estradiol level. METHODS: The experiment was performed at the Department of Anatomy, Histology and Embryology, Medical College of Hebei Engineering University from August 2006 to May 2007. (1)Seventy-two Sprague-Dawley female rats, 56-60 days old and 200-240 g, were selected. Sudan Ⅲ was product of Tianjin Kermel Reagent Factory. (2)The rats were randomly divided into ovariectomized group, sham-operation group, and normal group (n =24). After 2, 4, 6, and 8 weeks of feeding, 2% fluorescein sodium was dropped into rat conjunctival saccus and breakup time of tear film was observed by cobalt blue light scanning under slit lamp microscope. Then the rats were executed after anesthesia. (3)Serum estradiol level of rats was detected by means of radioimmunoassay. The positive staining and intensity of lipid in the meibomian gland alveolus was detected by Sudan Ⅲ staining. The experimental procedure was accorded with animal ethical standards. RESULTS: (1)The serum estradiol level was significantly decreased in ovariectomized group compared with normal groups and sham-operation group 4, 6 and 8 week after operation (P 〈 0.05, P 〈 0.01, P 〈 0.01). (2)Breakup time of tear film were significantly shortened in the ovariectomized groups than that the normal groups and sham-operation group 4, 6 and 8 week after operation (P 〈 0.05). (3)Sudan Ⅲ staining intensity was decreased significantly in the ovariectomized groups than that the normal groups and sham-operation groups 6 and 8 week after operation (P 〈 0.05, P 〈 0.01). (4)Serum estradiol level was strongly correlated with breakup time of tear film and Sudan Ⅲ staining intensity (correlation coefficient: r =0.995 7, P =0.004 3; r =0.972 3, P =0.003 6). CONCLUSION: Decrease in serum estradiol level after ovariectomy leads to reduce of lipid quality of the meibomian gland acinar epithelium, which is one of causes for the dysfunction of meibomian gland.
