高产辅酶Q_10光合细菌的选育 被引量:6

摘要 以球形红假单胞菌1.1737T为出发菌株,经紫外线和亚硝基胍诱变,并结合罗红霉素抗性和耐前体性物质-对羟基苯甲酸抗性突变株的筛选,获得1株辅酶Q10高产菌株PHBr-2,该菌株经发酵108 h的生物量为15.7g/L,辅酶Q10产量达到50.6mg/L,比原始菌株在同样条件下产量提高了90.9%。该变异株遗传性能稳定,经转接5代后其辅酶Q10产量为49.5mg/L,是1株很有前景的辅酶Q10生产菌株。 Rhodopseudomonas sphaeroides 1.1737^T was used as an original strain in this study R. sphaeroides 1.1737^T was mutagenized by UV and nitrosoguanidine (NTG) treatment. Then, a high coenzyme Q10-producting mutant R. sphaeroides PHB^r-2 was screened according to the tolerance on roxithromycin and precursor p-hydroxyben-zoic acid (PHB). Using R. sphaeroides PHB^r-2 for fermentation, the biomass reached 15.7 g/L at 108 h, and the coenzyme Q10 yield was 50.6 mg/L ( 90.9% higher than that of the parent strain). R. sphaeroides PHB^r-2 was genetic stable and produced 49.5 mg/L coenzyme Q10 after five-generation subculture. It was indicated that R. sphaeroides PHBr-2 was a good candidate for coenzyme Q10 pro duction.
