Peptidyl-prolyl isomerase 1:a catalytic molecule for oncogenesis and a potential therapeutic target

摘要 肽基脯氨酰异构酶1(peptidyl-prolyl isomerase1,Pin1)能特异性地催化磷酸化的丝/苏-脯氨酸基序发生异构,改变底物蛋白的构象,调节底物蛋白的功能。Pin1含有两个结构功能域:一个是N端的色氨酸-色氨酸中心区WW,另一个是C端的肽基脯氨酰异构酶PPlase活性区。Pin1在非肿瘤组织中呈低表达或无表达,但在多种肿瘤组织中呈过表达,并受到以E2F(腺病毒E2启动子相关细胞转录因子)为主的转录水平的调节。Pin1作用于多种底物如p53、c-Jun、cyclinD1等,通过催化异构改变底物的结构和功能,促进细胞增殖和肿瘤形成,阻断它的表达可以抑制肿瘤细胞的增殖,促进调亡。
