摘要
目的:借助扫描电镜观察,探讨垂体腺苷酸环化酶激活肽(PACAP)保护血管内皮的形态学证据。方法:雄性新西兰家兔60只,随机等分为动脉粥样硬化模型组(AS组)和PACAP干预组(P组)。于实验的第0、2、4、8、12周采集兔血清,测总胆固醇(TC)、甘油三酯(TG);于第0、4、8、12周处死每组兔各5~6只,取其主动脉,观察血管内膜面及内皮形态的改变。结果:2周时,两组兔血脂均已显著升高;至12周时,P组的TC、TG均显著低于同期AS组水平(P<0.05)。扫描电镜观察发现,随高脂血症的出现,AS组兔血管内膜出现损伤,且程度逐渐加重。早期可见明显的白细胞黏附,及内皮细胞的变形、隆起,接着可见内皮细胞破损、脱落,形成内膜面的"溃疡样"缺损。P组出现类似上述变化的时间较AS组晚,且病变的程度较同期AS组为轻。结论:PACAP可延缓并减轻高脂血症对内皮造成的形态损伤,具有明显的内皮形态保护作用。
Objective : By means of observation of scanning electron microscope ( SEM ), to explore the direct morphologic evidences of cytoprotective effects of pituitary adenylate cyclase-activating polypeptide(PACAP) on vascular endothelium.
Methods:Sixty male New Zealand rabbits were randomly divided into 2 groups:atherosclerosis model group (AS group)and PACAP group(P group), At the 0^th ,2^nd ,4^th ,8^th and 12^th week of the experiment,the concentrations of serum total cholesterol ( TC ) and triglyceride ( TG ) were measured. At the 0^th,4^th, 8^th and 12^th week, five to six rabbits in each group were sacrificed, the aorta were collected, and the pathological changes of intima and endothelium were observed by naked eyes and light microscope as well as SEM.
Results :The concentrations of both TC and TG in two groups were increased at 2^nd week. By the 12^th week of the experiment, the concentrations of TC and TG in P group were lower than those in AS group significantly( P 〈 0. 05 ). Under SEM, aorta in AS group were seen to be damaged due to hyperlipidemia, and the degree of damage became deterioration with time. At the early stage, the adhesion of white blood cells and the distortion and hunch of endothelial cells (ECs)were observed, followed by the dis- repair and shedding of ECs to form the ulcerous damages of the intima. But the similar damages of the aorta in P group were observed to occur late, and the damages are much weaker than those in AS group significantly.
Conclusion:PACAP can protect the vascular ECs against the injury induced by hyperlipidemia.
