摘要
目的:建立一个基于连接酶检测反应(LDR)的基因多态性检测系统,探讨降钙素基因相关肽(calcitoningene-relatedpeptide,CGRP)和白介素-1A(Interleukin-1A,IL-1A)基因多态性与中国汉族人群重度慢性牙周炎遗传易感性的关系。方法:收集100例重度慢性牙周炎病人和118例健康对照组的颊黏膜拭子并抽提DNA,应用PCR-LDR方法检测CGRP1210、CGRP4218、CGRP+5247以及IL-1A-889基因型,并使用软件SHEsis进行单体型分析,进行各组间基因型分布和等位基因频率的χ2检验。结果:重度慢性牙周炎病人组CGRP1210、CGRP+5247以及IL-1A-889阳性基因型均显著高于健康对照组(P<0.05),CGRP4218的基因型、等位基因频率在各组间的分布无显著性差异(P>0.05)。结论:建立了一个基于连接酶检测反应的基因多态性并行检测系统,IL-1、CGRP基因多态性与重度慢性牙周炎有相关关系,IL-1A-889、CGRP1210和CGRP+5247阳性基因型可能是重度慢性牙周炎易感性的遗传标志,同时提示IL-1、CGRP基因型在不同地域、不同人种中的分布可能是不相同的。
To establisha parallel typing system based on ligase detection reaction (LDR), and to study the correlation between CGRP and IL - 1A gene polymorphism and the susceptibility to severe chronic periodontitis in Chinese Han nationality. METHODS: Buccal swabs were collected from 100 adult patients with severe chronic periodontitis and 118 healthy adults. DNA was extracted from each sample. PCR -LDR technique was used to identify CGRPI210, CGRP + 5247, CGRP4218 and IL - 1A -889 genotype. The data were analyzed by χ^2 test using SHEsis. RESULTS: The results showed a significant difference in the gene type of IL - 1A -889, CGRPI210 and CGRP + 5247 between patients with severe chronic periodontitis and periodontally healthy controls. CONCLUSION: The parallel typing system based on ligase detection reaction was established for gene polymorphism research. There is a possible role of IL - IA and CGRP gene polymorphisms in the susceptibility to severe chronic periodontitis for some patients.
