摘要
目的:研究发现内脂素在多种代谢性疾病患者体内出现升高或高表达。实验旨在构建高脂饮食诱导胰岛素抵抗大鼠模型,并观察脂肪细胞因子内脂素在血清中的变化。方法:实验于2007-06/11在广州体育学院运动生化省重点实验室进行。①实验动物:将24只雄性SD大鼠随机分为对照组和高脂组,每组12只。实验过程中对动物处置符合动物伦理学要求。高脂饲料配方:猪油15%、白糖20%、蛋黄粉5%、胆盐0.2%、维生素0.05%、氯化胆碱0.15%、矿物质0.2%、基础饲料59.4%。②实验方法:适应性饲养1周后,分别喂以普通饲料和高脂饲料。6周末,禁食12h,麻醉后,腹主动脉取血,检测两组大鼠空腹血糖和空腹血胰岛素水平,用胰岛素抵抗指数模型评价胰岛素抵抗造模是否成功,并测定血清内脂素的浓度。分离附睾及肾脏周围脂肪垫,称质量,计为内脏脂肪总质量。结果:24只大鼠均进入结果分析。①6周末高脂组空腹胰岛素水平、胰岛素敏感指数和胰岛素抵抗指数高于对照组(P<0.01,P<0.05)。②6周末高脂组大鼠内脏脂肪总质量高于对照组(P<0.01)。③高脂组大鼠血清内脂素水平高于对照组(P<0.01)。结论:高脂饮食成功诱导胰岛素抵抗大鼠模型。胰岛素抵抗大鼠血清内脂素浓度明显增加,提示血清内脂素在胰岛素抵抗的发生和发展过程中可能起一定的作用。
AIM: Studies reveal that visfatin is increased or highly expressed in the patients with various metabolic diseases. This study was designed to establish the high fat diet (HFD)-induced insulin resistance (IR) model in rats, and explore the plasma level changes of adipocytokine visfatin. METHODS: The experiment was carried out in the Provincial Laboratory of Sports Biochemistry, Guangzhou Institute of Physical Education between June and November in 2007. Twenty-four SD male rats were randomly divided into HFD and control group equally. All the animals were handled according to the ethical standard. HFD prescription: lard 15%, white sugar 20%, yolk powder 5%, bile salt 0.2%, vitamin 0.05%, choline chloride 0.15%, mineral matter 0.2%, and basic forage 59.4%. After one-week adaptive feeding, rats in control group were fed with regular diet and those in HFD group were fed with HFD. Six weeks later, abdominal aorta blood samples were harvested after the rats were anesthetized, then the measurement of fasting blood glucose and fasting plasma insulin, meanwhile the IR index was evaluated to verify the modeling success. The plasma level of visfatin was checked in each group Fat of epididymis and fat pad of kidney were isolated as visceral fat, which would be weighed as visceral fat mass. RESULTS: All of 24 rats were involved in the result analysis. At the end of the sixth week, the fasting plasma insulin, insulin sensitivity index and IR index were all significantly high in HFD group than in control group (P 〈 0.01, P 〈 0.05). After 6-week feeding, the rats in HFD group had a increase in visceral fat mass and the plasma level of visfatin compared with control group (P 〈 0.01). CONCLUSION: IR model is successfully induced by HFD. Rats with IR have significantly increased the plasma level of visfatin, which suggests the possible role of visfatin performed in the occurrence and development of IR.
