肥大细胞对慢性实验性哮喘小鼠气道反应性和气道炎症的影响

摘要 目的研究肥大细胞在慢性实验性哮喘小鼠气道病理改变过程中的作用。方法遗传性肥大细胞缺失W/Wv小鼠、正常+/+小鼠和培养骨髓肥大细胞重建的W/Wv小鼠分设对照组和实验组。实验组小鼠用10μgOVA 2次致敏,17 d后用1%OVA气道激发,每周1次,共7周,检测其肺组织中肥大细胞数量、气道反应性和肺组织的病理改变。结果OVA致敏和激发可引起+/+和肥大细胞重建的W/Wv小鼠肺组织中肥大细胞数量明显增加,气道反应性增强,肺组织炎性病理改变明显;而OVA致敏和激发的W/Wv小鼠的上述指标改变不明显。结论肥大细胞参与了OVA诱导的小鼠气道高反应性、肺组织炎症细胞浸润过程。 Objective To investigate the role of mast cells in the pathophysiology of a chronic asthma model. Methods Genetical mast cell deficient W/W^v mice,their congenic normal littermates +/+ mice and BMCMC→W/^v mice were randomly divided into control and experimental groups;experimental mice were sensitized with 10 μg OVA,2 times;17 days after the beginning of the sensitization the mice received their airway challenges with 1% OVA weekly ;7 weeks later, their lung mast cell infiltration, airway reactivity, and lung pathologic change were studied. Results OVA sensitization and airway challenge could remarkably increase the number of lung mast cells, the airway reactivity, and the lung pathologic inflammatory change in the +/+ and BMCMC→W/W^v mouse; but the changes in OVA sensitized and challenged W/W^v mice were unconspicuous. Conclusion Mast cells take part in the airway high reactivity and lung inflammation in the chronic OVA-induced murine asthma.
