摘要
以青蒿组培苗幼嫩顶芽为材料,研究了秋水仙素诱导染色体倍性变异的最佳浓度和最适处理时间,结果表明:用0.2%秋水仙素溶液处理其试管苗幼嫩顶芽24 h的诱导方法最有利于四倍体的产生,诱导率达到36%。显微镜观察变异植株根尖染色体数目为2n=4x=36条。苗床生长2个月后,四倍体植株农艺性状较二倍体有明显的优越性。
Autotetraploid lines of Artemisia annua L were induced with colchicine. The results indicated that the optional methord for inducing autotetraploid was to immerse buds in 0.2 % colchicines solution for 24 h. Induced polyploids were identified by root-tip chromosome determination, and the result showed that their chromosome number was 2n=4x=36. The results of autotetraploid observation of the young plants in the seedbed showed that the autotetraploid materials were usually characterized by their higher plants, bigger leaves and thicker stems as compared with the diploid.
