栀子高频植株再生体系相关因子的优化 被引量:1

Optimization of Correlation Factors of High Efficiency Regeneration System of Gardenia Jasminoides Ellis.

摘要 采用正交实验和单因子实验以及植物组织培养的方法,对栀子高频植株再生体系有关因子的进一步优化进行了探讨。结果表明,栀子带芽茎段再生体系的最佳消毒方式为70%酒精处理30 s后再用0.1%HgCl2处理15 min;栀子带芽茎段再生的最佳培养基为MS+6-BA 2 mg/L+NAA 0.5 mg/L;光照培养有利于栀子带芽茎段的再生;栀子带芽茎段再生的最佳蔗糖浓度为25 g/L,最佳琼脂浓度为7.5 g/L,最佳pH值为6,最佳培养温度为25℃。 Objective: to illustrate the effects of several factors on dedifferentiation and redifferentiation of Gardenia jasminoides Ellis. earthnut. Method: orthogonal experiment, single factor experiment and tissue culture were applied. Result and Conclusion: the best disinfection way of Gardeniajasminoides Ellis. stems with a bud was firstly using 70% ethyl alcohol to process for 30 minutes then using 0.1%HgCl2 to process for 15 seconds again. The best regeneration culture medium of Gardeniajasminoides Ellis. stems with a bud was MS+6-BA 2 mg/L +NAA 0.5 mg/L. The light culture was more advantageous to Gardenia jasminoides Ellis. stem regeneration. For regeneration system of Gardenia jasminoides Ellis. stems with a bud, the best concentration sucrose and agar was 25 g/L and 7.5 g/L respectively, the best pH value was 6, the best culture tempreture was 25℃.
