摘要
目的探讨双花对人黑素瘤细胞增殖及黑素合成的影响。方法采用MTT法测定双花对人A375黑素瘤细胞活力的影响,以NaOH裂解法测定黑素合成,并与氢醌的结果进行比较。结果100μmol/L、500μmol/L和1000μmol/L双花对人A375黑素瘤细胞酪氨酸酶活性及黑素生成均有较强的剂量相关性抑制作用,500μmol/L及1000μmol/L双花与对照组相比有显著差异(P<0.01)。结论双花对于共培养体系酪氨酸酶活性及黑素生成有较强的剂量相关性抑制作用。
Objective To study the Effects of Lonicera japonica Thunb on Cen Proliferation and Melanin Synthesis of Cultured Human Melanoma Cells. Methods MTY method, NaOH - assay were employed to measure the cen viability, melanin synthesis of human A375 melanoma cens; tyrosinase activity was measured by utilization of L - Dopa as the substrate. Further-more, the effects of Lonicera japonica Thunb on human A375 melanoma cens were compared with that of hydroquinone. Resuits The melanin synthesis and tyrosinase activity were clearly suppressed by Lonicera japonica Thunb in a dose - dependent manner at concentrations of 100μM, 500μM, and 1000μM in human A375 melanoma ceils. 500μmol/L and 1000μmnol/L of Lonicera japonica Thunb showed statistically significant suppression ( p 〈 0.01 ) than the control. Conclusion Lonicera japonica Thunb exhibits a a dose - dependent inhibitory effects on melanin synthesis and tyrosinase activity in human A375 melanoma cells.
