血管内皮生长因子在喉粘膜上皮良性及恶性病变中的表达及定量 被引量:6

Quantitative observation of overexpression of vascular endothelial growth factor in benign and malignant epithelia of laryngeal mucosa

摘要 观察血管内皮生长因子(VEGF)在正常喉粘膜上皮、慢性非特异性炎症组织、不典型增生及鳞癌中的表达,用图像分析系统定量的方法观察VEGF与肿瘤的生长、浸润及转移的关系,观察VEGF是否可以作为一个判断预后的指标.方法:用抗VEGF抗体通过微波处理,暴露抗原,进行标准化的EliteABC染色,并通过图像分析系统确定VEGF在以上各组织中的表达.结果:VEGF在正常组织、炎症及不典型增生、鳞癌中均有表达,但在鳞癌组中的表达明显高于其它各组(P<0.05);在鳞癌组中发生转移的病变VEGF的表达明显高于未转移的鳞癌组(P<0.05);VEGF与病理分级无关,与临床分期密切相关(γ=0.5088,P<0.01).结论:VEGF不仅直接影响新生血管的形成,而且其表达与喉癌细胞的生长、浸润及转移密切相关,此外,肿瘤细胞也可表达VEGF进一步影响肿瘤细胞的生长和血管生成,VEGF可能作为一个判断预后的指标之一. To observe the overexpression of vascular endothelial growth factor (VEGF) in normal epithelia, chronic inflammation, dysplasia and malignant epithelia of laryngeal mucosa and quantify the immunoreactivities of VEGF by using the automated image analysis system, to observe the relationship between VEGF and the growth,infiltration,met astases in laryngeal carcinoma and prognosis value of VEGF. Methods:The overexpression of VEGF was determined using antibody against VEGF by standardized immunohistochemistry(Elite ABC method). Microwave oven heating was used for retrieval of antigen in paraffin-embedded tissues. An automated image analysis quantification was applied to determine the immunoreactivities of different lesions. Results: VEGF were expressed in all different lesions, and obviously higher in squamous cell carcinoma (SCC) lesions than in other lesions. The expression level of VEGF was higher in lesions with metastases than in the lesions without metastases. Related analysis indicated the overexpression of VEGF was correlated with the clinical stage, nor with the pathologic stage. Conclusion: The overexpression of VEGF is highly associated with the growth,infiltration and metastases of laryngeal carcinoma.
