摘要
目的建立一种测定家兔血浆中甲磺酸酚妥拉明含量的方法,并应用于甲磺酸酚妥拉明溶液的药动学研究。方法家兔的给药方式采用溶液灌胃;血药浓度分析采用HPLC内标法。结果甲磺酸酚妥拉明血药浓度测定的线性关系为:y= 0.006 6ρ-0.013 3(r=0.999 7,n=3),线性范围为:5~250μg·L^(-1)。6只家兔药动学参数分别为:ρ_(max)(110.78±24.35)μg·L^(-1),t_(max)(11.67±2.58)min,k(0.46±0.13)h^(-1),t(1/2)(1.60±0.47)h,AUC_(0→6h)(104.50±15.13)μg·h·L^(-1),AUC_(0→∞)(119.12±19.10)μg·h·L^(-1)。结论我们建立的测定方法简便、灵敏、准确,可以用作甲磺酸酚妥拉明在家兔体内的药动学研究。
AIM To establish a method to determine the concentration of phentolamine mesilate in rabbits, which can be applied to study the pharmacokinetics of phentolamine mesilate solution. METHODS A phentolamine mesilate solution was administered intragastrically to 6 rabbits. Shimadzu HPLC instrument was used to determine the plasma concentration. RESULTS The linear coefficient relation of phentolamine mesilate was y = 0.006 6·ρ- 0.013 3 (r = 0. 999 7, n = 3). The ρmax, t1/2, AUC0→6h and AUC0→∞ of phentolamine mesilate in rabbits were (110.78 ± 24.35)μg·h·L^-1, (11.67±2.58) min, (1.60±0.47) h, (104.50±15.13) μg·h·L^-1 and (119.12±19.10) μg·h·L^-1 respectively. CONCLUSION The method is practical and accurate to monitor the serum drug levels of phentolamine mesilate solution.
