摘要
目的建立快速经济有效提取微量外周血基因组DNA的方法。方法采用酚/氯仿提取法、NaCL沉淀法和试剂盒法,直接从人外周血中提取基因组DNA,并以此为模板,进行肿瘤坏死因子相关凋亡诱导配体(TRAIL)基因片段的扩增及鉴定。结果酚/氯仿法和试剂盒法提取的DNA量大,需血量少,用于PCR扩增结果稳定。结论酚/氯仿提取外周血DNA稳定、高效、经济,可用于批量临床标本的制备。
Objective To establish a rapid exist-effective method for extracting genomic DNA from human peripheral blood. Methods DNA was isolated from human peripheral blood by the methods of phonl/chloroform, NaCl's precipitation and kit. The DNA was used as template, then tumor necrosis factor related apoptosis inducing ligand (TRAIL) gene fragment was amplified and identificated by PCR. Results The genomic DNA extracted by methods of phenol / chloroform and kit was in large amount with less blood and the results of DNA amplification were stable. Conclusion Phenol / chloroform extraction is a stable, efficient and economical method, and it could be used to handle a large number of clinical smples.
