摘要
目的:观察三七总皂甙对佐剂性关节炎(AA)大鼠踝关节炎症和核因子-κB(NF-κB)表达水平的影响,探讨三七总皂甙治疗类风湿关节炎(RA)可能的作用机制。方法:24只雄性lewis大鼠随机分为正常对照组、模型组、三七总皂甙组和地塞米松组。大鼠右后足跖皮内注射弗氏完全佐剂建立AA模型,采用关节炎指数评估大鼠关节炎的情况,将大鼠踝关节病理切片分别做常规HE染色和免疫组化染色,对HE染色切片用光镜观察炎细胞浸润和滑膜细胞增生的情况,对免疫组化染色切片进行NF-κB表达的半定量分析。结果:三七总皂甙治疗2周后,大鼠多发性关节炎指数有所下降,与模型组比较差异有显著性(P<0.05),HE染色切片显示三七总皂甙组与模型组比较,关节炎症差异有显著性(P<0.05),免疫组化染色切片显示三七总皂甙组NF-κB表达下调,与模型组比较,差异有显著性(P<0.05)。结论:三七总皂甙有改善大鼠关节滑膜炎症,抑制NF-κB表达的作用,这可能是三七总皂甙治疗RA的作用机制之一。
Objective To observe the effects of panax notoginseng saponins (PNS) on ankle joint arthritis and expression of NF-KB in adjuvant arthritis (AA) rats and investigate the therapeutic mechanism of PNS in RA. Methods Twenty-four male lewis rats were randomly divided into four groups: normal control group, model group, PNS group and DM group. AA rat models were established by intradermal injection of Complete Freund's adjuvant. Polyarthritis indices were scored. Pathological sections of ankle joint in rats were made and then stained by HE method and immunohistochemical method respectively. Infiltration of inflammatory cell and hyperplasia of synovioblast in HE-stalned pathological sections were observed under microscope and expression of NF-κB was observed by semi-quantitative immunohistochemical method. Results PNS were given to rats by intraperitoneal injection for 2 weeks after immunization. Compared with AA model group, the polyarthritic symptoms of PNS group rats were improved significantly (P 〈 0.05), infiltration of inflammatory cell of PNS group rats was improved significantly in HE-stained pathological sections (P 〈 0.05 ) and expression of NF-κB of PNS group was decreased in pathological sections stained by immunohistochemical method (P 〈 0.05). Conclusion PNS has therapeutic effects on AA in rats, which may be related to improving arthrosynovitis and inhibiting expression of NF-κB.
