摘要
目的观察温脉通对血管平滑肌细胞(VSMC)增殖、迁移及其相关因子基质金属蛋白酶-2(MMP-2)和其抑制因子(TIMP-1)的影响。方法体外培养兔胸主动脉VSMC,以血管紧张素Ⅱ(AngⅡ)作为诱导因素,复制VSMC增殖、迁移模型,用温脉通药物血清进行干预。应用四唑盐(MTT)比色法测定VSMC增殖活性,用免疫细胞化学检测方法观察MMP-2、TIMP-1的表达。结果细胞经AngⅡ作用后其增殖活性和迁移距离明显增加,与正常对照组比较有显著性差异(P<0.01);而加入药物血清后,细胞增殖活性降低、迁移距离变短,与AngⅡ组比较有显著性差异(P<0.01),其中以高剂量组最为明显。正常对照组MMP-2呈弱表达、TIMP-1呈中度表达;AngⅡ组MMP-2表达明显增强、TIMP-1表达减弱;温脉通组MMP-2表达明显受到抑制而TIMP-1表达增强。结论温脉通可抑制AngⅡ诱导的VSMC增殖和迁移,抑制MMP-2蛋白表达、诱导TIMP-1蛋白表达,提示该方抑制VSMC迁移可能与影响细胞因子表达从而调节细胞外基质(ECM)代谢有关。
Objective To observe the effect of Wenmaitong on the proliferation and migration of vascular smooth muscle cell ( VSMC), and on the expressions of its correlation factors-matrix metalloproteinase-2 (MMP-2) and tissue inhibitor of metalloproteinase-1 (TIMP-1). Methods The model of VSMC proliferation and migration was established taken angiotensin Ⅱ (AngⅡ) as inducing factor in VSMC of rabbit aorta thoracalis cultured in vitro. The model was intervened by Wenmaitong medicated serum. The VSMC proliferation activity was determined by using methyl thiazolyl tetrazolium test ( MTT), and the expressions of MMP-2 and TIMP-1 were observed by using immunohistochemistry. Results Compared with the normal control group, the proliferation activity and migration of VSMC were increased in the AngⅡ group. There was a significant difference (P 〈 0.01 ). Compared with the AngⅡ group, the proliferation activity and migration of VSMC were decreased after treated with the medicated serum in the Wenmaitong group. There was a significant difference (P 〈0.01 ), and the difference was the most significant between the AngⅡ group and the large-dose Wenmaitong group. In the normal control group the expression of MMP- 2 was weak and expression of TIMP-1 was medium. In the AngⅡ group the expression of MMP-2 was increased and expression of TIMP-1 was decreased. In the Wenmaitong group the expression of MMP-2 was inhibited significantly and the expression of TIMP-1 was increased. Conclusion Wenmaitong can inhibit the proliferation and migration of VSMC induced by AngⅡ and the expression of MMP-2. It can also induce the expression of TIMP-1. The result indicates that the inhibitory effect of Wenmaitong on VSMC proliferation may be related to its influence on the cytokine expression and regulation to the metabolism of extracellular matrix.
