硒化壳聚糖对NB_4细胞的作用及其与NF-κB信号分子的关系 被引量:2

摘要 目的:研究硒化壳聚糖对人急性早幼粒细胞性白血病NB4细胞抗氧化指标的影响及其与核因子κB(NF-κB)信号分子的关系。方法:取NB4细胞加入不同浓度的硒化壳聚糖(50、100、200mg·L-1)作用24h后取上清检测谷胱甘肽过氧化物酶(GSH-px)、超氧化物歧化酶(SOD)、丙二醛(MDA)、NF-κB蛋白含量或活性,并设培养液为对照组。结果:与对照组比较,硒化壳聚糖各浓度组GSH-px活性明显升高(P<0.01),SOD活性和MDA含量明显降低(P<0.01或P<0.05),NF-κB蛋白含量明显降低(P<0.01),并呈量效关系。结论:硒化壳聚糖可通过减少进入细胞核内NF-κB蛋白含量的方式下调其下游基因表达,以及增强细胞抗氧化能力的方式抑制NB4细胞增殖,诱导细胞凋亡。 OBJECTIVE: To study the effect of selenium chitosan on anti- oxidative ability in NB4 cells in patients with acute promyelocytic leukemia (APL) and its correlation with the NF-κB signaling molecules. METHODS: NB4 cells were treated with different concentrations of selenium chitosan (50, 100, 200 mg · L^-1) for 24 h before the supernatant was taken for the determination of levels of GSH- px, SOD, MDA, and NF-κB signal protein molecules, with culture solution set as control. RESULTS: Compared with the control group, exposure of NB4 cells to different concentrations of selenium chitosan for 24 h could significantly increase GSH- px activity(P 〈 0.01), decrease SOD activity and MDA content(P〈0.01 or P〈0.05), and decrease the level of NF-κB in obvivous doseeffect relationship(P〈0.01) . CONCLUSION: Selenium chitosan downregulates gene expresxion of its downstream by reducing the level of NF-κB entering into nucleus and inhibits the proliferation of NB4 cells and induces apoptosis by enhancing the anti- oxidative ability of cells.
