摘要
目的观察邻苯二甲酸二异辛酯(DEHP)对雄性小鼠主要脏器及生殖功能的影响。方法将5~6周雄性小鼠24只随机分为4组,每组6只,对照组不做任何处理,低剂量组皮下注射DEHP 100mg·kg^-1,中剂量组皮下注射DEHP300mg·kg^-1,高剂量组皮下注射DEHP500mg·kg^-1;每131次,连续20d,停止给药3d后全部颈椎脱臼处死。测定小鼠体质量、主要脏器重量、精子活动率、精子存活率及精子畸形率。结果低剂量组、中剂量组小鼠肝脏系数与对照组比较差异显著(P〈0.05),高剂量组小鼠肝脏系数与对照组比较差异极显著(P〈0.01)。低剂量组、中剂量组、高剂量组的精子活动率和存活率随剂量的增加有明显下降趋势。低剂量组、中剂量组、高剂量组畸形数目随剂量的增加逐渐增多。结论DEHP能明显引起小鼠肝脏及生殖功能受损。
Objective To investigate the effect of di-(2-ethylhexyl) phthalate (DEHP)on main organ and reproductive function of male mice. Methods Twenty-four male mice were randomly divided into 4 groups,6 mice for each group. Control group do nothing. Low dosage group,middle dose group and high dose group were injected DEHP 100 mg · kg^-1 ,300 mg · kg^-1 and 500 mg · kg^-1 with hypodermic, respectively. Once two day for 20 days. On the third day after stopping giving medicine, killd the mice with cervical dislocation. The weight of mice and main organ, the reactivity and survival ratio of spermatozoa, the rate of teratospermia were detected. Results Compared with Control group, there was difference of liver coefficient in low dose group and middle dose group (P 〈 0.05 ), and there was significantly difference in high dose group( P 〈 0.01 ). The reactivity and survival ratio of spermatozoa decreased obviously when the dose of di-(2-ethylhcxyl)phthalate increased. The number of teratospermia was higher when the dose of di-(2-ethylhcxyl) phthalate increased. Conclusion DEHP can cause the damage of rat' s liver and reproductive function.
