摘要
目的观察快速老化痴呆鼠SAMP10痴呆状况与大脑总胆固醇异常的相关性,并探讨"益气调血、挟本培元"针法改善痴呆的机制。方法设立正常老化鼠SAMR1对照组,SAMP10对照组、针刺组及非穴组,通过Morris水迷宫测试各组小鼠学习记忆功能,Folch法和比色法测量大脑总胆固醇的含量。结果与SAMR1对照组相比,SAMP10对照组小鼠出现明显的学习记忆障碍,其大脑总胆固醇含量明显增加,差异有统计学意义(P<0.05);而针刺组可降低大脑总胆固醇含量,改善其学习记忆能力与SAMP10对照组比较差异亦有统计学意义(P<0.05)。结论调节大脑胆固醇的稳态可能是"益气调血、扶本培元"针法改善SAMP10鼠痴呆状况的机制之一。
Objective To observe the relationship between the brain total cholesterol and the dementia of senescence accelerated mice-prone 10 (SAMP10) and explore the mechanism of acupuncture method "reinforcing qi and regulating blood, strengthening the root and build up the strong constitution" in improving the dementia. Methods The Senescence-accelerated mice-resistant 1 (SAMR1) control group, SAMP10 control group, SAMP10 acupuncture group; and SAMP10 non-point group were established. The Morris water maze was used to test the learning ability and memory of mice of all groups, and the Folch and colorimetric methods were applied to observe the contents of their brain total cholesterol. Results Compared with the SAMR1 control group, the mice of SAMP10 control group had significant impairment in learning ability and memory and their brain total cholesterol was evidently increased, the difference was significant (P〈0.05). And the difference in the SAMP10 acupuncture group in decreasing the brain total cholesterol and improving the learning ability and memory was also significant in comparing with the SAMP10 control group (P〈0. 05). Conclusion Regulating the stability of brain cholesterol may be one of the mechanisms of acupuncture in "reinforcing qi and regulating blood, strengthening the root and build up the strong constitution" to treat dementia.
