摘要
目的建立妇炎康灌肠剂中小檗碱和丹酚酸B的含量测定方法。方法采用RP-HPLC法,色谱柱:Kromasil C_(18);流动相:乙腈-0.04 mol/L KH_2PO_4(含0.1%的H_3PO_4)(梯度洗脱);流速:1 mL/min;检测波长:230 nm;柱温:30℃。结果小檗碱和丹酚酸B分别在0.092 4~2.310 0μg和0.100 0~2.500 0μg范围内线性关系良好,r均为0.999 9。小檗碱高、中、低3个浓度平均加样回收率分别为100.40%,99.51 %,99.15%,RSD分别为0.59%,0.67%,1.02%;丹酚酸B高、中、低3个浓度平均加样回收率分别为101.52%,99.32%,98.68%,RSD分别为0.60%,0.92%,1.00%。结论该方法简便,专属性、重复性好,可用于妇炎康灌肠剂中小檗碱和丹酚酸B的含量测定。
Objective To establish a method for the determination of berberine and salvianolic acid B in Fuyankang En- ema. Methods The chromatograhic separation was performed on a Kromasil C18 column (250 mm × 4.6 mm, 5 μm) with a gradient elution of acetonitrile - 0.04 mol/L potassium dihydrogen phosphate solution containing 0. 1% phosphoric acid. The flow rate was 1.0 mL/min and column temperature was set at 30 ℃. The detection wavelength was 230 rim. Results There was a good linearity in the range of 0. 0924 - 2. 3100 μg ( r = 0. 999 9) for berbeine, and 0. 1000 - 2. 5000 μg ( r = 0. 999 9) for salvianolic acid B. The average recoveries of berberine at high, middle and low concentra- spectively. The average recoveries for salvianolic acid B were 101.52 % , 99. 32 % and 98.68 % respectively with RSD being 0. 60 % , 0.92 % and 1.00 % ( n = 3), respectively. Conclusions This method is convenient, specific and re- producible for the determination of berbeine and salvianolic acid B in Fuyankang Enema by HPLC.
