摘要
采用正向遗传学方法,从T-DNA标签技术构建的拟南芥突变体种子库中筛选到1株高叶绿素荧光表型的突变体(Hcf-1)。对该突变体及其子代群体进行Basta抗性及PCR分子检测,证实该突变体是由T-DNA插入引起的。突变性状与T-DNA共分离。对突变体(Hcf-1)和野生型(WT)叶片中色素含量、叶绿体内囊体膜色素蛋白复合物及蛋白亚基变化的分析结果表明,与野生型相比,突变体叶中的Chla和Chlb含量显著降低。光系统II部分解聚,其主要蛋白亚基CP43,CP47,D1,D2蛋白以及外周捕光色素天线蛋白LCHII含量均下降。光系统I基本未受到影响。据此判断由于T-DNA的插入,导致一个控制PSII发育的基因发生突变,叶绿体的发育和PSII的组装均受到较大的影响。
According to forward genetics approaches, a photosynthesis mutant of high-chlorophyll-fluorescence phenotype has been isolated by screening the seeds of T-DNA Transformed Activation Tag lines. Test for Basta resistance to the mutant and its generations showed the mutants were all resistant, which indicated that the phenotype was co-segregated with resistance, and T-DNA insertion was further confirmed using PCR method. The pigments content and the components of chloroplast protein complexes were investigated by BN-PAGE and SDS-urea-PAGE. The results showed: the content of pigments decreased significantly in Hcf-1. The contents of PSII dimer,PSII monomer,CP43- less PSⅡ and LHCⅡ monomer decreased. The contents of some crucial small molecular proteins CP43,CP47,D1,D2 and LHCⅡ monomer which are components of photosystem Ⅱ were degraded, whereas the content of reaction centre protein psa A/B were little affected. From the results above we deduced that the mutant location may lie in PSⅡ.
