摘要
以陕西部分地区分离的强致病性大肠埃希菌O2、O8、O135、O139、O141为研发菌种,通过菌种在不同的保存方法、温度、时间下保存后细菌的形态、存活率、菌落形态及对小鼠致病性的观察,最终确定了以含150mL/L牛奶蔗糖为保护剂的菌种,以冻干的方法于-20℃保存2年,以150mL/L甘油保护剂的菌种,于-20℃保存1年,两种保存方式和4℃可保存90d。
Several high pathogenic Escherichia coli stains isolated from some pig farms in Shaanxi province were preserved at different methods, temperature conditions, observed by colony morphology, livability of the bacteria and the pathogenicity to mice after some time. The results indicated that the bacteria main tained viability and pathogenicity for 2 years when stored at the optimal preservation condition at -20 ℃ and lyophilized in 15% milk-sucrose or mixed with 15% glycerol for 1 year at -20 ℃, ninety days at 4 ℃ in above two methods.
