摘要
目的:探讨ERK-1mRNA在CCl4致实验性肝纤维化大鼠肝组织中的表达及姜黄素(curcumin)对它们的影响。方法:建立CCl4诱导的大鼠实验性肝纤维化的模型,用免疫组化法比较正常组、模型组以及姜黄素组大鼠的α-平滑肌肌动蛋白(α-smooth muscle action,α-SMA)的表达,用RT-PCR法比较各组大鼠肝组织胞外信号调节激酶-1(Extracellular-regulated kinase-1,ERK-1)的表达。结果:模型组大鼠α-SMA以及ERK-1mRNA的表达增高明显,而姜黄素组则较模型组低。结论:姜黄素可能通过抑制α-SMA的表达,抑制ERK-1的促肝星状细胞(hepatic stellate cell,HSC)的增殖作用,从而产生抗四氯化碳诱导的大鼠肝纤维化的作用。
Objective: To evaluate Expression of ERK-1mRNA in rat with live,' fihrusis and the influence of cureumin to them. Methods: Liver fibrosis in rats were induced by carbon tetrachloride (CCl4) . Rats were divided into three groups: norreal control group, CCl4 model group and cureumin group. The expression of alpha-Smooth Muscle Action were detected by immunohistochemistry. The mRNA expression of extraeetlular signal-regulated kinase (ERK-1) were determined by RT- PCR. Results: The expression of alpha-Smooth Muscle Action were significantly h,wer in the rats trealed with eureumin compared with those of the model group ( P 〈0. 05) . The mRNA expression of ERK-1 were signitieantly higher in model group, but obviously lower in cureumin treated group (P 〈0.05 ) . Conclusion: cureumin is able to inhibit the aetiviation of HSC through suppress ing the expression of alpha-Smooth Muscle Action and it also can inhibit the proliferation of HSC through suppressing the mRNA expression of ERK-1.
