摘要
建立应用高效液相色谱法测定发酵液中武夷菌素含量方法。色谱柱ZORBAX SB-AQ(4.6mm×250mm i.d.,5μm);柱温25℃;流动相为1.4g/L三氯乙酸水溶液,流速1mL/min;检测波长254nm。将色谱峰面积与武夷菌素质量浓度进行线性回归,表明在0.2~1.6g/L质量浓度范围内线性关系良好,变异系数为0.43%~2.08%,回收率为96.59%~100.57%。
A high performance liquid chromatography (HPLC) method for the determination of wuyiencin of an agricultural antibiotic in fermentation broth was developed. Analytical conditions involved use of a SB-AQ column (4.6 mm × 250 mm i.d., 5 μm) with temperature of 25 ℃. The mobile phase was 1.4 g/L trichloroacetic acid,flow rate was 1 mL/min and detection wavelength was 254 nm. It also showed a good linearity within the range of 0.2-1.6 g/L. The recovery rate was 96.59-100.57% and the relative standard deviation was 0.43-2.08%.
