摘要
建立紫外分光光度法测定γ-聚谷氨酸含量的方法。利用γ-聚谷氨酸水解成谷氨酸,在pH9.6硼砂缓冲溶液中,能与四氯对苯醌形成1:1的络合物,该络合物在波长350nm处有最大吸收,通过测定其吸光度确定γ-聚谷氨酸含量。结果表明,γ-聚谷氨酸在8~12μg/mL范围内线性关系良好(r为0.9981),平均回收率为99.5%,相对标准偏差为1.0%。该方法简便准确、重现性好,可作为γ-聚谷氨酸含量的测定方法。
This paper establishes an ultraviolet spectrophotometric method for determination of γ-polyglutamic acid. After hydrolysis γ- polyglutamic acid became aminoglutaminic acid which could shape comoles compound (1 : 1) in pH 9.6 of borax buffer solution with chloranil. The comoles compound had absorption maximum at 350 nm. Thus we determined γ-polyglutamic acid by utilizing its absorbance. There was good linearity in the range of 8-12 μg/mL of γ- PGA (r=0. 998 1) and the average recovery was 99.5% with RSD of 1.0 %. The method is simple, reliable and reproducible which can be used to assay γ - PGA.
