摘要
目的研究氢溴酸加兰他敏凝胶剂中加入氢溴酸东莨菪碱前后的透皮特性。方法采用改良的Franz扩散池,分别以离体SD大鼠皮肤和新西兰家兔皮肤为渗透屏障,HPLC测定接受液中氢溴酸加兰他敏与氢溴酸东莨菪碱的含量,比较单方与复方的透皮速率。结果氢溴酸加兰他敏凝胶中加入氢溴酸东莨菪碱,2种成分的含量测定不相互干扰(P>0.05),且透皮速率发生改变。氢溴酸加兰他敏单独与复方后透过鼠皮的速率分别为43.88,86.67μg.cm-2.h-1,透过兔皮的速率为26.49,75.49μg.cm-2.h-1;氢溴酸东莨菪碱单独与复方后透过鼠皮的速率分别为104.66,170.96μg.cm-2.h-1,透过兔皮的速率分别为17.87,75.93μg.cm-2.h-1。结论氢溴酸加兰他敏与氢溴酸东莨菪碱复方后增加彼此的透皮速率,有利于发挥这2种药物的协同作用。
OBJECTIVE To study the permention characteristics of gels contained scopolamine hydrobromide and/or galantamine hydrobromide in vitro. METHODS The penetrating experiments were performed by using rabbit skin and rat skin respectively on modified Franz diffusion cell. The contenls of scopolamine hydrobromide and galantamine hydrohronfide were determined by HPLC. RESULTS The determination drugs showed no interference with each other(P 〉 0. 05). The permention flux through rat skin of scopolamine hydrobromide in the different gels with scopolamine hydrobromide done and coexisted with galantamine hydrobromide were 104. 66, 170.96 μg·cm^-2·h^-1, respectively. The permeation rate through rabbit skin were 17.87,75.93μg·cm^-2·h^-1. The permention flux through rat skin of galantamine hydrobromide in the different gels with scopolamine hydrobromide done and coexisted with seopolamine hydrobromide were 43.88,86.67 μg·cm^-2·h^-1. The permeation rate through rabbit skirl were 26.49,75.49 μg·cm^-2·h^-1. CONCLUSION The permention flux in vitro will he enhanced by scopolamine hydrobromide coexisted with galantamine hydrobromide in gel.
