摘要
基质金属蛋白酶(MMPs)和表皮生长因子(EGF)与肿瘤的发生和转移有密切关系,为了研究EGF对MMP-12的调节机制,我们用不同浓度的EGF处理人表皮癌细胞(A431细胞)后,观察MMP-12的表达变化。结果显示使用不同浓度EGF处理A431细胞后,MMP-12的mRNA和蛋白水平均有所升高。因此,在A431细胞中,EGF通过上调MMP-12的表达可能会促进肿瘤的侵袭和转移。
Matrix metalloproteinase（MMPs） and epidermal growth factor （EGF） are both have a close relationship with tumor＇ s genesis and metastasis. To investigate the effects of EGF on MMP-- 12 in human epidermoid carcinoma A431 ceils, we treated A431 cells with different concentration of EGF to observe the changes of MMP--12＇s expression. After the A431 cells treated with EGF, the mRNA and protein expression of MMP--12 were both increased. Along with the hoist of the EGF＇s concentration, the expression of MMP--12 was augment. In the process of tumor metastasis, EGF could regulate the expression of MMP--12 to promote the tumor＇s invasion. Therefore, the high expression of MMP--12 could accelerate the degradation of extracelluar matrix, and probably provide an proper condition for the tumor＇s transfer.
