摘要
目的通过观察IFN-γ对于体外培养的肝星状细胞(HSC)分泌细胞外基质的影响,探讨IFN-γ在治疗肝纤维化中的作用机制。方法采用体外培养大鼠肝星状细胞(rHSC-99),加入不同剂量的IFN-γ,并用放射免疫分析法测定分泌PCⅢ、LN、HA含量的改变。结果IFN-γ对rHSC-99细胞分泌PCⅢ、LN、HA的抑制作用随着用药剂量的增大而明显增强。随培养时间的延长,IFN-γ对rHSC-99细胞分泌PCⅢ、LN、HA的抑制作用逐渐增强。除HA外,IFN-γ的浓度和培养时间与rHSC-99细胞分泌PCⅢ、LN值均呈显著负相关。结论IFSI-γ对rHSC-99细胞分泌PCⅢ、LN、HA具有抑制作用,具有浓度—效应依赖关系、时间-效应依赖关系。
AIMS: The purpose of this research is to present IFN-γ's function mechanisms of the anti-fibrosis through observing the influence of IFN-γ on the vitro HSC's extracellular matrix (ECM) secretion, and to provide further theoretical foundations for treatment of liver fibrosis in clinic. METHODS: HSC were cuhured in vitro,and were treated with IFN-γ. The levels of PC Ⅲ, LN and HA wereexamined by radiommunoassy (RIA) . RESULTS: The impact of IFN-γon the secretions of Pc m, LN, HA will be stronger with a higher dose. IFN-γ significantly reduces PCⅢ, LN, HA secreting when the culturing time is extended. Except for HA, the levels of PC Ⅲ and LN are negatively correlated with the dose and culturing time of IFN-γ. CONCLUSION: IFN-γ has a negative effect on the secretion of ECM by rHSC-99, and exists concentration-effect relationship and tine-effect relationship.
