摘要
目的观察硫辛酸对阿霉素所致小鼠心肌损伤的保护效应。方法把75只小鼠随机分成5组:正常对照组,阿霉素损伤组和低、中、高剂量硫辛酸组。正常对照组腹腔注射生理盐水10ml/kg;阿霉素损伤组腹腔注射阿霉素3mg/kg,隔日1次,连用7次;低、中、高剂量硫辛酸组腹腔注射阿霉素3mg/kg,隔日1次,连用7次,于实验第4天每天腹腔注射硫辛酸10mg/kg、20mg/kg、40mg/kg,连用10d,实验结束后检测相关指标。观察小鼠精神状况和存活率,心肌超氧化物歧化酶活力,心肌丙二醛含量,血清诱导型一氧化氮合酶含量和还原型谷胱甘肽含量。结果①存活率:阿霉素损伤组低于阿霉素+高、中、低剂量硫辛酸组。②心肌丙二醛含量和血清诱导型一氧化氮合酶含量:阿霉素损伤组明显高于正常对照组(P<0.01),高、中、低剂量硫辛酸组明显低于阿霉素损伤组(P<0.01)。③心肌超氧化物歧化酶活性和血清还原型谷胱甘肽含量:阿霉素损伤组明显低于正常对照组(P<0.01),高、中、低剂量硫辛酸组明显高于阿霉素损伤组(P<0.01)。结论硫辛酸可明显减轻由阿霉素所致小鼠心肌损伤的程度。硫辛酸的作用机制可能与其清除氧自由基及抗脂质过氧化等作用有关。
Objective To observe the protective effect of α-lipoic acid against adriamycin-induced cardiotoxicity in mice. Methods Totally 75 mice were randomly divided into five groups:normal control group, adriamycin injured group and three dosage groups.The normal control group was treated with saline 10 ml·kg^-1·d^-1 by intraperttoneal injection. The adriamycin injured group was treated with 3 mg/kg adriamyein by intraperitoneal injection , once every two days for seven times. The α-lipoic acid groups were established in the same way of adriamycin injured group with α-lipoic acid 10 mg/kg, 20 mg/kg and 40 mg/kg daily intraperitoneal injection for 10 days on the fourth day of the experiment . Relevant indexes were measured after the experiment was finished. The health conditions were observed. The activities of plasma superoxide dismutase (SOD) were measured .The contents of reduced glutathione(GSH) and inducible nitricoxide synthase(iNOS) in blood serum were measured respectively. Results (1)The survival rate of the model group was lower than the normal control group and groups. (2)The contents of serum MI3A and contents of iNOS in blood serum were significantly higher in the adriamycin injured group than the normal control group and three dosage groups (P〈0.01). (3)The activities of plasma SOD in myocardial tissues of mice and contents of GSH were significantly lower in the adriamycin injured group than the normal control group and three dosage groups (P〈0.01). Conclusion α-lipoic acid significantly alleviates the degree ofthe myocardial injury in mice induced by adriamycin. Its mechanism may be associated with reducing oxygen free radical production and anti-lipoperoxidation.
