摘要
目的研究柴胡皂苷a(SSa)对谷氨酸(G lu)激活大鼠海马星形胶质细胞内Ca2+浓度([Ca2+]i)和白介素-6(IL-6)释放的影响。方法将原代培养并纯化的海马星形胶质细胞分为4组:对照组、L-G lu激活组(含L-G lu 0.1 mmol/L)和含L-G lu 0.1 mmol/L+SSa的药物干预组(SSa剂量分别为1.25 mg/L和0.625 mg/L),作用12 h后,运用Fluo3/Am荧光法测定星形胶质细胞内[Ca2+]i,ELISA法测细胞外液IL-6水平。结果G lu可显著升高星形胶质细胞[Ca2+]i(P<0.01),并使细胞外液IL-6水平显著增加(P<0.01);SSa能明显抑制G lu激活星形胶质细胞([Ca2+]i)的增加(P<0.01),并能降低G lu激活星形胶质细胞外液中IL-6的水平(P<0.01);其中1.25 mg/L剂量的SSa对G lu激活的星形胶质细胞细胞内[Ca2+]i升高和IL-6释放均有较好的抑制作用,与对照组比较,无显著性差异(P>0.05)。结论SSa可抑制G lu激活的星形胶质细胞[Ca2+]i升高和IL-6释放,这可能是其抗癫痫作用机制之一。
Objective To study the effect of saikosaponin a (SSa) on intracellular free calcium concentration ( [ Ca^2+ ] ) and interleukin-6 in hippoeampal astroeytes actived by glutamate. Methods A cell culture model of neonatal Spague-Dawley rat hippocampal astrocytes were used. After exposed to extracellular fluid with calcium, L-glutamate (0. 1 mmol/L ), L-glutamate and SSa of different concentration (1.25 mg/L and 0. 625 mg/L) for 12 hours, changes of [ Ca^2+ ]i and IL-6 were observed by enzyme-linked immunosorbent assay (ELISA) and fluorescent Ca^2+ indicator Fura-3/AM respectively. Results Elevation of intracellular Ca^2+ fluorescence intensity and increased expression of IL-6 (P 〈 0.01 ) were observed in glutamate group. Compararing with glutamate group, elevation of [ Ca^2+] i and IL-6 expression were decreased after astrocytes were incubated with SSa ( P 〈 0.01 ). The [Ca^2+]i and IL-6 level in 1. 25 mg.L^-1 of SSa group and control group showed no significant differences (P 〉 0.05 ). Conclusion SSa could effectively inhibit the hippocampal astrocytes from decreasing [ Ca^2+ ]i. and secreting IL-6, and the inhibitory effect on elevation of [ Ca^2+ ]i and IL-6 expression may be one of the mechanisms of its anti-epilepsy effect.
