脑心康片对大鼠脑缺血再灌注损伤后Caspase-3 mRNA表达的影响 被引量:1

摘要 目的:观察脑心康片对大鼠脑缺血再灌注损伤的保护作用,并进一步探讨其作用机制。方法:Wistar大鼠雄性60只,按照体质量随机分为6组,即脑心康片高剂量组、脑心康片中剂量组、尼莫地平组、银杏叶片组、模型组和假手术组,每组10只。分别给等体积生理盐水,各组均连续灌胃给药5 d后,采用大鼠大脑中动脉线栓法制备不完全脑缺血模型,RT-PCR法检测脑心康片对脑缺血后脑组织Caspase-3 mRNA表达的影响。结果:脑心康片能明显抑制Caspase-3 mRNA的表达。结论:脑心康片对脑缺血具有保护作用,其机理可能与脑心康片通过抑制凋亡相关基因的表达,进而抑制神经元细胞凋亡有关。 Objective: It was studied and explored further the mechanism of treatment that the protective effect of phenolic alkaloids from Menispermum dauricum(PAMD) on cerebral ischemia in rats. Methods: In order to test the protective effect of PAMD treating cerebral ischemia, the rats with incomplete cerebral ischemia were induced with fishing line in the middle cerebral artery occlusion (MCAO). It was detected the gene expression of Caspase-3 mRNA with RT-PCR. Results: PAMD could inhibit expression Caspase-3 mRNA. Conclusion: PAMD inhibited cell apoptosis and affected the related gene expression interrelated with cell apoptosis.
